產(chǎn)品描述
PU.1-Luc熒光素酶報告基因（報告基因質(zhì)粒）（PU.1 luciferase reporter plasmid）是翊圣生物自主研發(fā)的用于檢測PU.1轉(zhuǎn)錄活性水平為目的的報告基因。PU.1轉(zhuǎn)錄因子因其DNA結(jié)合區(qū)識別共有序列GAGGAA，故該區(qū)又被稱為PU.1 box。PU.1主要在造血系統(tǒng)如髓細胞和B淋巴細胞中表達，調(diào)節(jié)關(guān)鍵髓系基因的轉(zhuǎn)錄從而調(diào)控造血系統(tǒng)的分化。
PU.1報告基因主要用于檢測細胞中PU.1的活性、藥物研究以及基因過表達和RNAi的表型分析等。
pGMPU.1-Lu是翊圣生物改造后的哺乳動物真核表達載體，在其多克隆位點插入了多個PU.1結(jié)合位點，可以高靈敏度地檢測PU.1的激活水平。同時，對載體中預(yù)測出的其它轉(zhuǎn)錄因子以外的結(jié)合位點進行了適當?shù)耐蛔?，增加了質(zhì)粒的轉(zhuǎn)錄因子結(jié)合特異性。由于質(zhì)粒體積減小，使得PU.1報告基因更易于轉(zhuǎn)染。

使用說明
pGMPU.1-Lu可以采用常規(guī)轉(zhuǎn)染方法轉(zhuǎn)染哺乳動物細胞。用熒光素酶檢測試劑盒或雙熒光素酶檢測試劑盒進行檢測。
