hPSMA/MC-38
| I. Background | |||
| PSMA 蛋白也稱為羧基肽酶 II 和水解酶 1，它是一種跨膜糖蛋白酶，存在 于細(xì)胞表面。PSMA 主要在前列腺、腎臟、小腸以及中樞和外周神經(jīng)系統(tǒng)中表達(dá)。PSMA 在正常前列腺組織和非前列腺癌組織（如淚腺、神經(jīng)系統(tǒng)、十二指腸）中表達(dá)水平很低， 但在前列腺癌組織中的表達(dá)比正常組織高 100-1000 倍。在前列腺癌中，PSMA 表現(xiàn)出羧肽 酶活性，可以促進(jìn)釋放，并激活系統(tǒng)，調(diào)節(jié)鈣離子通道。此外，它能夠 將細(xì)胞生存信號從絲裂原激活蛋白激酶途徑轉(zhuǎn)向磷酸-3 激酶/Akt 致癌途徑，最終促進(jìn) 腫瘤細(xì)胞生長。PSMA 在其他實(shí)體腫瘤中也有表達(dá)，比如胃癌、胰腺癌、乳腺癌、肺癌等， 但是在這些腫瘤中，PSMA 分布于腫瘤血管而不是腫瘤細(xì)胞。在非前列腺癌中，PSMA 主 要參與腫瘤血管的生成，PSMA 可促進(jìn)血管粘連蛋白肽的下游 MMP-2 的激活。 | |||
