研究背景：IL-15R α 在多種細胞類型上表達，包括淋巴細胞、骨髓細胞、非淋巴細胞和非造血細胞[4]。IL-15R α 在 Epstein-Barr 病毒相關性胃癌 (EBVaGC) 中通過啟動子高甲基化下調[5]。 IL-15R α 的氨基酸序列在不同細胞中存在差異。物種。人 IL-15R α 與小鼠的氨基酸序列同一性 IL-15R α 是將 IL-15 從內(nèi)質網(wǎng)轉運到細胞表面以與 β (CD122) 和 γ (CD132) 結合所必需的) 響應淋巴細胞上的鏈[4][6]。當與 IL-15 結合時，該復合物可延長 IL-15 的體內(nèi)半衰期，并增強 NK 細胞和 CD8+ T 細胞中 IL-15 與 IL-15Rβ/γ 的結合親和力。因此，信號傳遞可提高 NK 細胞和 CD8+ T 細胞的增殖和抗腫瘤活性[2]。此外，癌細胞表面的 IL-15R α 會誘導惡性表型，例如癌細胞生長、遷移和侵襲增強，細胞凋亡減少[5]。 IL-15R α 結合與 IL-15 結合，激活 NK 細胞和 CD8+ T 細胞的抗腫瘤功能，對記憶 CD8 T 細胞穩(wěn)態(tài)和淋巴細胞發(fā)育也很重要[2][3] 。
