（1）取河沙加入10％稀鹽酸，加熱煮沸30分鐘，以去除其中的有機(jī)質(zhì)。
（2）倒去酸水，用自來(lái)水沖洗至中性。
（3）烘干，用40目篩子過(guò)篩，以去掉粗顆粒，備用。
（4）另取非耕作層的不含腐植質(zhì)的瘦黃土或紅土，加自來(lái)水浸泡洗滌數(shù)次，直至中性。
（5）烘干，碾碎，通過(guò)100目篩子過(guò)篩，以去除粗顆粒。
（6）按一份黃土、三份沙的比例（或根據(jù)需要而用其他比例，甚至可全部用沙或全部用土）摻合均勻，裝入10×100mm的小試管或安瓿管中，每管裝1g左右，塞上棉塞，進(jìn)行滅菌，烘干。
（7）抽樣進(jìn)行無(wú)菌檢查，每10支沙土管抽一支，將沙土倒入肉湯培養(yǎng)基中，37℃培養(yǎng)48小時(shí)，若仍有雜菌，則需全部重新滅菌，再作無(wú)菌試驗(yàn)，直至證明無(wú)菌，方可備用。
（8）選擇培養(yǎng)成熟的（一般指孢子層生長(zhǎng)豐滿的，營(yíng)養(yǎng)細(xì)胞用此法效果不好）優(yōu)良菌種，以無(wú)菌水洗下，制成孢子懸液。
（9）于每支沙土管中加入約0．5ml（一般以剛剛使沙土潤(rùn)濕為宜）孢子懸液，以接種針拌勻。
（10）放入真空干燥器內(nèi)，用真空泵抽干水分，抽干時(shí)間越短越好，務(wù)使在12小時(shí)內(nèi)抽干。
（11）每10支抽取一支，用接種環(huán)取出少數(shù)沙粒，接種于斜面培養(yǎng)基上，進(jìn)行培養(yǎng)，觀察生長(zhǎng)情況和有無(wú)雜菌生長(zhǎng)，如出現(xiàn)雜菌或菌落數(shù)很少或根本不長(zhǎng)，則說(shuō)明制作的沙土管有問(wèn)題，尚須進(jìn)一步抽樣檢查。
（12）若經(jīng)檢查沒(méi)有問(wèn)題，用火焰熔封管口，放冰箱或室內(nèi)干燥處保存。每半年檢查一次活力和雜菌情況。
（13）需要使用菌種，復(fù)活培養(yǎng)時(shí)，取沙土少許移入液體培養(yǎng)基內(nèi)，置溫箱中培養(yǎng)。
