Luciferase報(bào)告基因系統(tǒng)是以熒光素(luciferin)為底物來檢測螢火蟲熒光素酶(fireflyluciferase)活性的一種報(bào)告系統(tǒng)。然后可以通過熒光測定儀也稱化學(xué)發(fā)光儀(luminometer)或液閃測定儀測定luciferin氧化過程中釋放的生物熒光。熒光素和熒光素酶這一生物發(fā)光體系，可以極其靈敏、高效地檢測基因的表達(dá)。是檢測轉(zhuǎn)錄因子與目的基因啟動(dòng)子區(qū)DNA相互作用的一種檢測方法。轉(zhuǎn)錄因子是一種具有特殊結(jié)構(gòu)、行使調(diào)控基因表達(dá)功能的蛋白質(zhì)分子，也稱為反式作用因子。某些轉(zhuǎn)錄因子僅與其靶啟動(dòng)子中的特異序列結(jié)合，這些特異性的序列被稱為順式作用元件，轉(zhuǎn)錄因子的DNA結(jié)合域和順式作用元件實(shí)現(xiàn)共價(jià)結(jié)合，從而對基因的表達(dá)起抑制或增強(qiáng)的作用。熒光素酶報(bào)告基因?qū)嶒?yàn)（luciferase assay）是檢測這類轉(zhuǎn)錄因子和其靶啟動(dòng)子中的特異順序結(jié)合的重要手段。
其原理簡述如下：
（1）構(gòu)建一個(gè)將靶啟動(dòng)子的特定片段插入到熒光素酶表達(dá)序列前方的報(bào)告基因質(zhì)粒，如pGL3-basic等。
（2） 將要檢測的轉(zhuǎn)錄因子表達(dá)質(zhì)粒與報(bào)告基因質(zhì)粒共轉(zhuǎn)染293細(xì)胞或其它相關(guān)的細(xì)胞系。如果此轉(zhuǎn)錄因子能夠激活靶啟動(dòng)子，則熒光素酶基因就會(huì)表達(dá)，熒光素酶的表達(dá)量與轉(zhuǎn)錄因子的作用強(qiáng)度成正比。
