| I. Background | |
| DLL4（Delta-like ligand 4） 主要表達(dá)于血管內(nèi)皮細(xì)胞，通過 “細(xì)胞-莖細(xì)胞”分 化機(jī)制調(diào)控血管分支：DLL4 激活鄰近細(xì)胞的 Notch1 信號，抑制其分化為細(xì)胞，從而 限制血管過度生長，維持血管網(wǎng)絡(luò)的有序性。在腫瘤微環(huán)境中，DLL4 在異常血管中高表 達(dá)，誘導(dǎo)血管結(jié)構(gòu)紊亂（滲漏、低灌注），促進(jìn)缺氧和耐藥。此外，DLL4 參與神經(jīng) 發(fā)育及 T 細(xì)胞分化，其基因突變可導(dǎo)致 Adams-Oliver 綜合征等先天性疾病。 DLL4 與 Notch1 是 Notch 信號通路 的關(guān)鍵組分，通過細(xì)胞間接觸依賴性信號調(diào)控發(fā)育、 血管生成及疾病進(jìn)程。DLL4 是 Notch 配體家族成員，作為跨膜蛋白特異性地結(jié)合并激 活 Notch1 受體，驅(qū)動(dòng)細(xì)胞命運(yùn)決定、組織穩(wěn)態(tài)及病理重塑。 | |
